The Effect of Zinc on Alkaline Phosphatases”

چکیده
Zinc has been shown to be an element essential to the life and growth of rats, but as yet there is no clear indication of what its function may be in metabolism. There are several reports of the occurrence of zinc in pure biological compounds. Scott and Fisher (1) have shown zinc to be necessary for the crystallization of insulin, although other metals were also effective. Keilin and Mann (2) report that carbonic anhydrase contains 0.32 per cent zinc. However, it has been well established that this enzyme is inhibited by small amounts of added zinc (Meldrum and Roughton (3), and unpublished data from this laboratory). In connection with the role of this red blood cell enzyme it may be significant that a zinc-porphyrin compound is excreted in the urine of humans (4). A zinc-porphyrin has also been isolated from diptherial toxic broth (5). Foster and Waksman (6) have shown zinc to be essential for the growth and fumaric acid production of Rhizopus nigricans. Recently Holmberg (7) has found that a purified uricase preparation contains 0.13 per cent zinc. In previous work from this laboratory (8, 9) we have been able to find no detectable difference in the carbohydrate metabolism of zinc-deficient rats. However, a disturbance of the nitrogen metabolism was observed. This involved a decreased blood nonprotein nitrogen, a marked delay in the rate of absorption of amino acids fed by mouth, an increase in fecal nitrogen excretion, a decrease in the daily urinary creatinine excretion, and a marked

Application of alkaline phosphatases from different sources in pharmaceutical and clinical analysis for the determination of their cofactors; zinc and magnesium ions.
Prospects of using different alkaline phosphatases bearing zinc and magnesium ions in their catalytic and allosteric sites, respectively, in pharmaceutical and clinical analysis were demonstrated. Also their application for the determination of zinc in insulin to control injection quality and magnesium in human urine for the diagnosis and treatment of magnesium deficiency was shown. The reactio...
متن کاملThe biosynthesis of apo- and metalloalkaline phosphatases of Escherichia coli.
A system for detecting the effects of metal ions, repressors, and mutations on the biosynthesis in vivo of alkaline phosphatase of Escherichia coli is described. If E. coli constitutive for alkaline phosphatase are grown in a medium containing 65Zn and are subject to osmotic shock, about 50% of the zinc taken up by the cells is released into the medium. Chromatography of this medium shows that ...
متن کاملEffect of plant growth hormones and abiotic stresses on germination, growth and phosphatase activities in Sorghum bicolor (L.) Moench seeds
Phosphatases are widely found in plants having intracellular and extracellular activities. Phosphatases are believed to be important for phosphorous scavenging and remobilization in plants, but its role in adaptation to abiotic stresses and growth hormones at germination level has not been critically evaluated. To address this issue, the effect of ABA, GA3, NaCl and drought on germination, grow...
متن کاملMicrobial alkaline phosphatases in bioprocessing
Microorganisms producing alkaline phosphatases are wide spread in nature. Alkaline phosphatase (Orthophosphoric monoester phosphohydrolase, E.C.3.1.3.1) is a hydrolase enzyme functioning at alkaline pH. The metalloenzyme containing the two zinc ions are involved in catalysis whereas the magnesium ion is important in structural stabilization (Simpson et al., 1968; Anderson et al., 1975). The enz...
متن کاملNucleotide sequence of the gene for alkaline phosphatase of Thermus caldophilus GK24 and characteristics of the deduced primary structure of the enzyme.
The gene encoding Thermus caldophilus GK24 (Tca) alkaline phosphatase was cloned into Escherichia coli. The primary structure of Tca alkaline phosphatase was deduced from its nucleotide sequence. The Tca alkaline phosphatase precursor, including the signal peptide sequence, was comprised of 501 amino acid residues. Its molecular mass was determined to be 54¿ omitted¿760 Da. On the alignment of ...
متن کاملDifferential Expression of Arabidopsis thaliana Acid Phosphatases in Response to Abiotic Stresses
The objective of this research is to identify Arabidopsis thaliana genes encoding acid phosphatases induced by phosphate starvation. Multiple alignments of eukaryotic acid phosphatase amino acid sequences led to the classification of these proteins into four groups including purple acid phosphatases (PAPs). Specific primers were degenerated and designed based on conserved sequences of PAPs isol...
